شناسایی مولکولی اورنیتوباکتریوم راینوتراکئال در پرندگان حیات وحش استان خوزستان

چکیده

اورنیتوباکتریوم راینوترکئال (ORT) به‌عنوان یکی از ارگانیسم‌های دخیل در بیماری‌های تنفسی گونه‌های مختلف پرندگان شناخته شده‌است. تاکنون گزارش‌های متعددی مبنی بر جداسازی این باکتری از گله‌های صنعتی پرورش طیور در ایران ارائه شده‌است. پژوهش حاضر با هدف شناسایی مولکولی و تعیین حساسیت آنتی‌بیوتیکی باکتری ORT در پرندگان حیات وحش استان خوزستان طراحی گردید. بدین‌منظور بعد از جمع‌آوری تعداد 23 نمونه سواب نایی، با استفاده از روش‌های مبتنی بر کشت، چهار (3/17 درصد) جدایه مظنون به باکتری ORT از این نمونه‌ها جداسازی گردید که با به‌کارگیری پرایمرهای اختصاصی، سه (13 درصد) جدایه تایید مولکولی شدند. تمام سویه‌های جداشده در این مطالعه از راسته شاهین ‌سانان (4/21 درصد) بودند که نشان‌دهنده میزان بالای آلودگی این پرندگان نسبت به دیگر طیور تحت بررسی می‌باشد (05/0 > p). بر اساس نتایج آزمایش تعیین الگوی مقاومت دارویی جدایه‌ها به روش انتشار دیسک، همه جدایه‌ها (100 درصد) نسبت به انروفلوکساسین، پنی‌سیلین، استرپتومایسین، سفالکسین، لینکواسپکتین، تایلوزین، فلورفنیکل، فلومیکوئین و تتراسایکلین حساس بوده و بیشترین مقاومت دارویی (6/66 درصد) نیز مقابل فسفومایسین، سولتریم، نالیدیکسیک اسید و جنتامایسین مشاهده گردید. در مقایسه با مطالعات انجام‌یافته بر روی سویه‌های جداشده از طیور صنعتی به‌خصوص ماکیان، سویه‌های شناسایی شده در پرندگان حیات وحش نسبت به اغلب عوامل ضدمیکروبی حساسیت نشان دادند.

Ornithobacterium rhinotracheale (ORT) is known as one of the involved organisms in respiratory diseases of different species of birds. So far, numerous reports have been presented about the isolation of the bacterium from commercial poultry flocks in Iran. This study was designed for molecular identification and evaluation of antimicrobial susceptibility of ORT bacterium in wildlife birds in Khouzestan province. After collection of 23 tracheal swab samples, four (17.3%) strains of ORT bacterium were isolated based on cultural methods; then three (13%) isolates were confirmed by application of specific primers. All strains isolated in this research were from Falconiformes (21.4%), which indicates the high rate of ORT isolation than that of other understudy birds (P<0.05). According to the results of determination of drug-resistance patterns of the isolates by disk diffusion method, all isolates (100%) were found sensitive to enrofloxacin, penicillin, streptomycin, cephalexin, lincospectin, tylosin, flumequine, florfenicol and tetracycline and 66.6% of them were resistant to fosfomycin, sultrim, nalidixic acid and gentamicin. In comparison with studies on strains isolated from commercial flocks especially poultry, strains detected in wild birds were sensitive to most antimicrobial agents.
