Iran has still been recognized as the major center for glanders. The present study was conducted to isolate B. mallei from blood specimen of an infected horse with glanders reported from Oshnavieh-West Azerbaijan in the past decade and molecular identification based on specific genes BimA, IS407-flip, and 23S rRNA.The blood sample of infected horse were cultured in biphasic medium containing nutrient broth and nutrient agar supplemented with glycerin and antibiotic. The bacterial isolate was identified by biochemical tests. The isolate was inoculated to male guinea pig intraperitoneally as a sensitive host for pathobiological studies. B. mallei isolate was verified by PCR and sequencing of BimA, IS407-flip, and 23S rRNA genes. B. mallei was isolated from blood sample of infected horse. The major sign of testicular swelling was seen in the male guinea pig after about 72 h IP inoculation with B. mallei and testicular colonization. The PCR amplification of BimA, IS407-flip, and 23S rRNA genes of B. mallei resulted in expected sizes of 989 bp, 250 bp, and 526 bp, respectively. Burkholderia pseudomallei and Pseudomonas aeruginosa were used as negative control.The report of glanders is alarming for healthcare organizations and need to monitor carefully. Due to the complication of diagnosis of glanders infectious agent, the identification of B. mallei using BimA, IS407-flip, and 23S rRNA is according to the diagnostic standards of World Organisation for Animal Health (OIE).}, keywords = {Burkholderia mallei,BimA,IS407-flip,glanders}, title_fa = {شناسایی و تعیین هویت مولکولی باکتری بورخولدریا مالئی جدا شده از نمونه‌ی خون اسب مبتلا به مشمشه}, abstract_fa = {بورخولدریا مالئی عامل اتیولوژیک بیماری زئونوزی مشمشه، یکی از خطرناک‌ترین و قدیمی‌ترین بیماری‌های واگیردار در تک‌سمیان است. کشور ایران به عنوان کانون مشمشه در دنیا شناخته شده است. مطالعۀ حاضر به‌منظور جداسازی باکتری بورخولدریا مالئی از نمونۀ خون یک مورد اسب مبتلا به مشمشه در منطقۀ اشنویه آذربایجان غربی در دهه گذشته و تعیین هویت مولکولی ارگانیسم با استفاده از ژن‌های اختصاصی BimA ،IS407-flip و 23S rRNA انجام گرفت. نمونۀ خون اسب مبتلا به مشمشه در محیط دی‌فازیک حاوی بویون و ژلوز گلیسیرینه همراه با آنتی‌بیوتیک کشت داده شد. بورخولدریا مالئی با آزمون‌های بیوشیمیایی شناسایی و خالص‌سازی شد. ایزولۀ باکتری با تزریق به میزبان حساس و بررسی علائم پاتوبیولوژیکی آن، تایید شد. تعیین هویت ایزولۀ کلینیکی بورخولدریا مالئی بر اساس تکثیر و تعیین توالی ژن‌های اختصاصی BimA ،IS407-flip و 23S rRNA باکتری انجام گرفت. بورخولدریا مالئی از نمونۀ خون اسب مبتلا به مشمشه جداسازی و شناسایی شد. حدود 72 ساعت متعاقب تزریق سوسپانسیون باکتری بصورت درون‌صفاقی به خوکچه هندی نر و کلونیزاسیون باکتری در بافت بیضه، علامت بارز تورم بیضه در حیوان مشاهده شد. تکثیر قطعات BimA ،IS407-flip و 23S rRNA محصولات مورد انتظار در اندازه‌های به ترتیب 989، 250 و 526 جفت باز را نشان داد. بورخولدریا پسودومالئی و پسودوموناس آئروژینوزا به عنوان کنترل منفی استفاده شدند. گزارش مشمشه هشداری برای مراقبت‌های بیشتر نهادهای بهداشتی است. با توجه به مشکلات تشخیص سریع و دقیق عامل عفونی مشمشه، شناسایی ارگانیسم با استفاده از ژن‌های اختصاصی BimA ،IS407-flip و 23S rRNA مطابق با استانداردهای تشخیصی سازمان جهانی بهداشت حیوانات (OIE) صورت گرفت.}, keywords_fa = {بورخولدریا مالئی,BimA,IS407-flip,مشمشه}, url = {https://vj.areeo.ac.ir/article_118544.html}, eprint = {https://vj.areeo.ac.ir/article_118544_ecc6b8dc3e6c870c2ce2f91e3738d1df.pdf} }
