In this search (2006-2009), isolation of Mycobacterium bovis, the causative agent of BTb,from tuberculin positive (reactor) cattle slaughtered in Isfahan has been addressed. Mesenteric and mediastinal lymph nodes along with pathological specimens(if present) from 143 cattle were cultured on Lownstein-Jenson (LJ) slopes. The 28 isolates representing mycobacterial growth characteristics eg. Acid fastness in microscopy, were subjected to a standard biochemical algorythm comprising preferential growth on pyruvate LJ medium, catalase, niacin, nitrate reductase, tiophen carboxylic hydrazide acid (TCH) and production of cord factor tests. Consequently, all the 28 isolates were identified as M. bovis. These isolates originated from 8 farms in different regions of Isfahan province with 15 of them coming from a single herd in Baharestan followed by two more farms with 4 isolates from each. The remaining 5 single isolates were collected from 5 herds in Dolatabad, Najafabad, Tiran, Golshar and Ghaleh Shour regions. Isolation of M. bovis as the single mycobacterial causative pathogen in the recator cattles of  Isfahan is another confirmatory observation that no other member of Mycobacterium tuberculosis complex plays a role, if any, in epidemiology of BTb in Isfahan as is the case with rest of Iran. In three out of the 8 farms studied here, there were 4 or more M. bovis isolates collected from every herd an indication of BTb epidemics in these herds. Whether single strain or multiples strains of M. bovis have been involved in the disease outbreaks in these farms, furher molecular epidemiology work is required. }, keywords = {Tuberculin positive,Tuberculusis,Mycobacterium bovis,Acid Fast,Reactor}, title_fa = {جداسازی و شناسایی مایکوباکتریوم بویس از نمونه‌های کشتارگاهی گاوهای توبرکولین مثبت استان اصفهان با استفاده از کشت میکروبی و تعیین هویت بیوشیمیائی}, abstract_fa = {علیرغم اجرای طولانی برنامه مبارزه با سل گاوی در ایران، این بیماری همچنان به عنوان یک مشکل بزرگ باقی مانده است و سبب ایجاد ضررهای اقتصادی گسترده می‌شود. در این مطالعه جداسازی مایکوباکتریوم بوویس، عامل ایجادکننده سل گاوی، از گاوهای رآکتور(توبرکولین مثبت) کشتارشده در استان اصفهان مورد بررسی قرار گرفته است. در فاصله زمانی 1385تا 1388 عقده‌های لنفاوی مزانتریک و مدیاستینال همراه با نمونه‌های پاتولوژیک (در صورت وجود) از143راس گاو بر روی محیط کشت اختصاصی لونشتاین- جانسون گلیسیرین دار و لونشتاین- پیرووات دار کشت داده شد. تعداد 28 جدایه دارای ویژگی‌های رشد مایکوباکتریوم‌ها نظیر برخورداری از خاصیت اسید فست بودن در میکروسکوپی، توسط یک الگوریتم استاندارد بیوشیمیایی متشکل از آزمایش‌های رشد ترجیهی بر روی محیط کشت بیرووات دار،کاتالاز، تولید نیاسین، احیاء نیترات، رشد در محیط حاوی تیوفن کربوکسیلیک هیدرازید اسید (TCH) و تولید کورد فاکتور، مورد بررسی قرار گرفتند. در نتیجه تمام 28 جدایه به عنوان مایکوباکتریوم بوویس شناسایی شدند. این جدایه‌ها از 8 واحد گاوداری در نواحی مختلف استان اصفهان جمع‌آوری شده بودند که از میان آنها 15 جدایه مربوط به یک گله در بهارستان و دو گله دیگر هر یک با 4 جدایه بودند. 5 جدایه باقیمانده از 5 گاوداری در مناطق دولت آباد، نجف آباد، تیران، گلشهر و قلعه شور اصفهان جداسازی شدند. جداسازی م.بوویس به عنوان تنها پاتوژن مایکوباکتریایی عامل بیماری در گاوان رآکتور اصفهان نشانه تاییدکننده دیگری است دایر بر اینکه دیگر اعضاء کمپلکس م.توبرکولوزیس نقشی در اپیدمیولوژی سل گاوی در این استان ندارند، همچنان که این مشاهده در مورد دیگر استان‌های ایران نیز موضوعیت دارد. در سه مورد از 8 گاوداری مورد بررسی در این بررسی 4 جدایه م. بوویس و یا بیش از آن از هر گله جمع‌آوری گردید که این امر نشانه وقوع همه‌گیری بیماری در این گله‌ها می‌باشد. پاسخ به این سؤال که آیا یک سویه واحد یا چندین سویه در بروز اپیدمی در این گله‌ها دخالت داشته اند نیازمند انجام بررسی‌های دقیق مولکولار اپیدمیولوژی خواهد بود.}, keywords_fa = {تست توبرکولین,مایکوباکتریوم بویس,تست های بیوشیمیایی,اسید فست,رنگ آمیزی ذیل نلسون}, url = {https://vj.areeo.ac.ir/article_106063.html}, eprint = {https://vj.areeo.ac.ir/article_106063_0231795e2736237a931300bd0767204a.pdf} }
