In order to maintain their potency and efficacy, stabilizer agents are usually added to the lyophilized vaccines during preparation process, stabilizing the vaccine throughout the manufacturing, storage, and administration. This study aimed to determine the efficacy of four different stabilizers on IBV. Four different stabilizers including lactalbomin–lactose (LL), lactalbumin–sucrose (LS), lactose–peptone (LP) and gelatin–sorbitol (GS) were used to prepare lyophilized vaccines. The vaccine was titrated before and after lyophilization. To determine accelerated stability (based on OIE instruction), the vaccines were titrated after 7 days incubation in 37ºC. Residual moisture of lyophilized vaccines was measured by Karl-Fisher method. Furthermore, the stability of the vaccines in 4ºC was measured over 36 months at three months intervals. All experiments were repeated for three batches of the vaccine. The results showed that acceptable titer of the vaccine was observed with four stabilizers. Residual moisture found to be in appropriate limit, ranged from 1.42-2.72%. Following seven days incubation in 37ºC, titer reduction of the vaccine with lactose-lactalbomin was the lowest. The results showed that in the end of this period, the vaccine with LL has the highest titer. In conclusion, it seems that lactose-lactalbomin stabilizer provided good protection to IBV under lyophilization condition and maintenance of the vaccine for long time in 4ºC.}, keywords = {Infectious bronchitis virus,vaccine,Stabilizer}, title_fa = {ارزیابی کارآیی پایدارکننده‌های شیمیایی مختلف بر روی پایداری واکسن برونشیت عفونی طیور}, abstract_fa = {واکسن برونشیت عفونی طیور (IBV) یک واکسن ویروسی زنده تخفیف حدت یافته بوده که نسبت به تغییر شرایط محیطی حساس می باشد. پایدارکنندهها به منظور حفظ توانمندی و کارآیی واکسن، قبل از لیوفیلیزه شدن به مایع واکسن اضافه می شوند که منجر به حفظ پایداری واکسن در طی مراحل تهیه، نگهداری و تلقیح می شود. این تحقیق به منظور تعیین کارآیی چهار ترکیب پایدارکننده بر پایداری واکسن IBV پس از لیوفیلیزه شدن انجام شد. چهار پایدارکننده لاکتوز- لاکتالبومین (LL)، سوکروز- لاکتالبومین (LS)، لاکتوز-پپتون (LP) و ژلاتین- سوربیتول (GS) برای تهیه واکسن لیوفیلیزه شده مورد استفاده قرار گرفت. واکسنهای فرموله شده با پایدارکنندهها پس از لیوفیلیزاسیون عیار سنجی شدند و با عیار اولیه مایع واکسن مقایسه گردید. سپس طبق دستورالعمل OIE، برای تعیین پایداری تسریع شده، عیار واکسنها بعد از یک هفته انکوباسیون درºC 37 تعیین گردید. رطوبت واکسن پس از لیوفیلیزاسیون با روش کارل- فیشر اندازه گیری شد. همچنین برای تعیین پایداری طولانی مدت واکسن در ºC 4، برای مدت 3 سال و هر سه ماه یکبار عیارسنجی شدند. آزمایشهای فوق برای 3 سری واکسن تولید شده انجام گردید. نتایج نشان داد واکسنهای لیوفیلیزه شده با پایدار کنندهها عیار قابل قبول را داشتند و میانگین رطوبت واکسن‌ها در حدود قابل قبول (72/2-42/1%) بود. پس از یک هفته انکوباسیون واکسنها در °C 37 (پایداری تسریع شده)، واکسن با پایدارکننده لاکتوز-لاکتالبومین کمترین کاهش تیتر را داشت. همچنین نتایج نشان داد که در پایان دوره سه ساله نگهداری واکسن‌ها در  ºC 4، واکسن با پایدارکننده LL بالاترین عیار را داشت. بدین طریق به نظر میرسد ترکیب پایدارکننده لاکتوز-لاکتالبومین حفاظت خوبی در طی مرحله لیوفیلیزاسیون و همچنین نگهداری طولانی مدت در  ºC 4 برای واکسن IBV فراهم مینماید.}, keywords_fa = {ویروس برونشیت عفونی,واکسن,پایدارکننده}, url = {https://vj.areeo.ac.ir/article_103031.html}, eprint = {https://vj.areeo.ac.ir/article_103031_1ecd7fc44805d5cdf619c2cf79a77950.pdf} }
